The mitochondrial TOM complex (TOM) is the central gateway for newly synthesized proteins passing through the mitochondrial outer membrane. This talk will discuss our latest 3.3 Å cryo-EM structure of the complex and a 4 Å map detailing its interaction with a precursor protein at the late stage of translocation. By constraining the complex’s lateral mobility through physical interaction in the membrane, I will show that TOM responds to mechanical interactions, not just to biochemical signals. TOM may thus be the first indication of mechanosensitivity within the beta-barrel porin family, in addition to the already known mechanosensitivity of alpha-helical proteins.
