DESCRIPTION:In cells, DNA methyltransferases (DNMTs) are targeted to genomic regions by several mechanisms. In this talk, I will present a novel assay allowing to explore DNMT specificities systematically, some of the results, and explain some of the molecular mechanisms causing the sequence specificity of DNMTs. I will also present recent results demonstrating that DNMTs methylate DNA in cells with strong preferences for flanking sequences. Hence, cellular DNA methylation patterns are modulated by the enzyme inherent flanking sequence preferences. However, it will be shown that many molecular mechanistic details of the DNMT-DNA interaction are still unknown.
